Should be checked the lift off supports in design temperature or operating temperature is enough?

Depends on your company standards although in my humble opinion you should not use design.

If doing stress checks, I would ensure stress passes under design conditions.

i think u should check at design temperature,
make hot load sustained case to see the primary stress is within limit or not
